Understanding clonal evolution using game theory and single-cell long-read isoform analysis


Understanding mechanisms that drive cellular differentiation has broad implications for a variety of fields including stem cell research, cancer research, and developmental biology.

Single-cell isoform sequencing with the new PacBio MAS-Seq for 10x Single Cell 3’ kit enables researchers to capture cryptic regulatory events that drive these differentiation processes such as alternative splicing (AS), alternative promoter regulation (APR), and alternative polyadenylation (APA), at single-cell resolution.

In this webinar, Dr. Nathan Salomonis of Cincinnati Children’s Hospital Medical Center presents how he used the MAS-Seq kit and sequencing on the PacBio Revio system to provide new insights into leukemia clonal evolution, through paired single-cell mutation, transcription, splicing, cell-surface, and cell state analysis. Learn how this methodology allowed for the characterization of unique lineages, an important step in single-cell transcriptomics and cancer research.
