In this technical note, we evaluate a modified “heat-then-extract” workflow that uses a lower temperature during the thermal annealing step, compared to the standard PacBio AAV protocol.
This approach may show improved recovery of full-length genomes while reducing partial snapback artifacts, with consistent results across Revio, Sequel IIe, and Vega platforms.
